The NF-E2 DNA binding protein is composed of two subunits, p45 and MafK, and it regulates expression of globin genes in developing erythroid cells through interaction with Maf recognition elements (MAREs). A family of NF-E2 related proteins, which are collectively known as the Cap 'n' collar (CNC) family and include Nrf1 (also designated TCF11), Nrf2 and Nrf3, are bZIP transcription factors that heterodimerize with Maf proteins to bind MARE sequences. The Nrf proteins also bind the antioxidant response element (ARE) and are implicated in the regulation of detoxification enzymes and the oxidative stress response. The major constituents of mucus, the viscous secretion that covers epithelial surfaces such as those in the trachea, colon, and cervix, are highly glycosylated proteins called mucins. These glycoproteins play important roles in the protection of the epithelial cells and have been implicated in epithelial renewal and differentiation. This gene encodes an integral membrane glycoprotein found on the cell surface, although secreted isoforms may exist. MUC-4 transcripts have been detected in normal respiratory epithelium and lung. MUC-4 is a very specific (100%) and sensitive (90%) marker of lung adenocarcinomas and is negative for mesotheliomas. Reportedly, MUC-4 expression in invasive ductal carcinoma of the pancreas is an independent factor for poor prognosis. Mammaglobin is a 93 amino acid glycoprotein with homology to other secretoglobin-uteroglobin family members. It was originally identified as a breast cancer restricted biomarker by differential screening. Mammaglobin related to secretoglobin family that includes human uteroglobin and lipophilin. Mammaglobin antibody stains cytoplasm of normal breast epithelial cells as well as primary and metastatic breast carcinomas. Mammaglobin expression is absent in prostate, kidney, colon, rectum, small intestine, stomach, pancreas, lung, and thyroid tissues. Spectrin is a cytoskeletal protein that lines the intracellular side of the plasma membrane of many cell types in pentagonal or hexagonal arrangements, forming a scaffolding and playing an important role in maintenance of plasma membrane integrity and cytoskeletal structure. In certain types of brain injury such as diffuse axonal injury, spectrin is irreversibly cleaved by the proteolytic enzyme calpain, destroying the cytosketelon. Spectrin cleavage causes the membrane to form blebs and ultimately to be degraded, usually leading to cell death. RNA polymerase II (Pol II) is an enzyme that is composed of 12 subunits and is responsible for the transcription of protein-coding genes. Transcription initiation requires Pol II-mediated recruitment of transcription machinery to a target promoter, thereby allowing transcription to begin. The largest subunit of Pol II (referred to as RPB1 or RPB205) is a 1, 840 amino acid protein that contains one C2H2-type zinc finger and a C-terminal domain comprised of several heptapeptide repeats. Although Pol II function requires the cooperation of all twelve subunits, the largest subunit conveys Pol II catalytic activity and, together with the second largest subunit, forms the active center of the Pol II enzyme. This MAb recognizes a protein of 63 kDa, identified as ROR-C. Its epitope maps in between aa1-50. The nuclear orphan receptors ROR and ROR are members of the nuclear hormone receptor superfamily. Members of this family act by directly associating with DNA sequences known as hormone response elements (HREs) and typically bind DNA as either homo- or heterodimers. RORalpha and RORgamma are unique in that they bind DNA as monomers. The C-type lectin superfamily member CD72 is a cell surface negative regulator of B cell activation from the pro-B through the mature B cell stage. CD72 serves as a receptor for CD5. Recognizes a 53 kDa protein, identified as the Ornithine Decarboxylase (ODC-1). ODC is the initial and rate-limiting enzyme in the biosynthetic pathway of polyamines and is involved in the conversion of ornithine to putrescine. The biological activity of ODC-1 is rapidly induced in response to virtually all agents known to promote cell proliferation including hormones, drugs, growth factors, mitogens, and tumor promoters.
